چکیده
loxiazine ring. Effect of pH on fluorescence of the flavin peptide was shown in fig. 1. The fluorescence of the flavin peptide was quenched at neutral and alkaline pH ranges. This shows that the amino acid linked to flavin ring is a histidyl residue.6) The pKa. value of the flavin peptide was found to be about 4.6, which was almost identical with that of the authentic 8a-[7V(3)-histidyl]riboflavin.

Design and properties of human D-amino acid oxidase with covalently attached flavin.
An "artificial flavinylation" approach was developed to replace a native noncovalent flavin prosthetic group with a covalently attached flavin analogue in recombinant human d-amino acid oxidase. The protein residue Gly-281 was replaced with Cys by site-directed mutagenesis, followed by reaction between mutated apoenzyme and the thiol-reactive flavin analogue, 8-methylsulfonyl FAD. The stoichiom...
متن کاملNew oxidase from Bjerkandera arthroconidial anamorph that oxidizes both phenolic and nonphenolic benzyl alcohols.
A new flavooxidase is described from a Bjerkandera arthroconidial anamorph. Its physicochemical characteristics, a monomeric enzyme containing non-covalently bound flavin adenine dinucleotide (FAD), and several catalytic properties, such as oxidation of aromatic and polyunsaturated aliphatic primary alcohols, are similar to those of Pleurotus eryngii aryl-alcohol oxidase (AAO). However, it also...
متن کاملThe d-Amino Acid Oxidase, Uricase, and Choline Oxidase in Two Transplanted Rat Tumors and in Isolated Nuclei of Tumor Cells*
In order to determine whether or not changes in enzymatic activity specifically accompany neoplasia, and to what extent such changes may occur, many enzymes have been investigated by Greenstein (2) in both normal and neoplastic tissues. Shack (3) has reported that d-amino acid oxidase is low in rat hepatoma 31, and suggested that the lowering of the enzyme may be owing to a deficiency in protei...
متن کاملBiochemical and Mechanistic Studies of Nitronate Monooxygenase and Roles of Histidine Residues in Select Flavoprotein Oxidases
Nitronate monooxygenase (NMO) catalyzes the flavin-dependent oxidation of propionate 3-nitronate (P3N) via the formation of an anionic flavosemiquinone. The oxidation of substrate includes the formation of a peroxy-nitro acid intermediate. P3N is activated to its radical form via a single electron transfer onto the FMN cofactor forming the anionic flavosemiquinone. Reoxidation of FMN cofactor f...
متن کاملAlcohol oxidase: a complex peroxisomal, oligomeric flavoprotein.
Alcohol oxidase (AO) is the key enzyme of methanol metabolism in methylotrophic yeast species. It catalyses the first step of methanol catabolism, namely its oxidation to formaldehyde with concomitant production of hydrogen peroxide. In its mature active form, AO is a molecule of high molecular mass (600 kDa) that consists of eight identical subunits, each of which carry one non-covalently boun...
